The determination of 17,21-dihydroxy-20-ketosteroids in urine and plasma.

چکیده
In 1950 (l), the authors described a color reaction between 17,21-dihydroxy-20-ketosteroids and phenylhydrazine and showed that, after addition of 1 r or more of cortisone per ml. of urine or plasma, about 88 per cent could be detected in chloroform extracts. Nelson and Samuels (2) improved the procedure, primarily by employing chromatographic separation and micro cuvettes, so that they were able to measure the concentration of hydrocortisone in human plasma. For routine clinical use, however, it seemed desirable to develop a relatively simple procedure which could be completed within a working day. This communication is concerned with a description of such a procedure, which involves four steps : (1) a 15 second extraction with 5 volumes of chloroform, (2) a 10 second washing with dilute alkali, (3) a 15 second extraction of an aliquot of the chloroform extract with the alcoholic phenylhydrazine-sulfuric acid reagent, and (4) measurement of optical density in micro cuvettes in the Beckman spectrophotometer after heating the samples to 60” for 30 minutes.

THE DETERMINATION OF URINARY 17-TRIOXYSTEROIDS* BY RAYMOND RIVOIRE,t
The color reaction characteristic of cortisone and related compounds (with the 17,21-dihydroxy-20-keto grouping), which Porter and Silber (1) reported in 1950, has been used by a number of investigators to measure the “cortisone-like” metabolites of urine. To facilitate expression, we propose to give to the methods of analysis utilizing this reaction the term of “determination of 17-trioxystero...

The numerous combinations of functional groups occurring in the steroid hormones render difficult the development of a specific test for individual compounds. However, reactions which reveal certain groupings are available and may be applied singly or in combination to elucidate the characteristics of pure steroids or of compounds in a mixture. The ketol side chain, located at carbon atom 17 in...
متن کاملThe effect of intravenously-administered ACTH on plasma 17,21-dihydroxy-20-ketosteroids in normal individuals and in patients with disorders of the adrenal cortex.
The response of the adrenal cortex to exogenous ACTH has been recognized as one of the more specific and quantitative tests of adrenal function (1). Changes in the 24-hour excretion values of urinary 17-ketosteroids and 17-hydroxycorticosteroids during ACTH administration have proved to be valuable indices of adrenocortical capacity (2, 3). With the development of technics for the chemical esti...

IN1938, Anderson, Haymaker, and Joseph (1) reported the finding of increased concentrations of cortin in the blood and urine of patients with Cushing’s syndrome. Since then, as methods for the determination of cortical steroids have been improved, they have been applied to the urine and plasma of patients as a measure of adrenocortical activity. The reaction of 17, 21-dihydroxy-20-ketosteroids ...
متن کاملThe influence of the thyroid on adrenal cortical function.
The evidence is quite conclusive from animal studies that an excess of circulating thyroid hormone results in adrenal enlargement, and hypothyroidism produced by surgical thyroidectomy or by administration of antithyroid drugs results in adrenal atrophy. This subject has been extensively reviewed by Money (1). Studies of adrenal size in patients with thyroid disease coming to autopsy have yield...
